Evodiamine

As we know protein level of SERT is drastically reduced after chronic administration of SSRI

"The aim of this study was to investigate the effect of berberine and evodiamine on serotonin transporter (5-HTT) expression and then test how allelic variations previously identified in the promoter region could modulate that effect in the serotonergic neuronal cell line RN46A. Both berberine and evodiamine, alone and in combination, increased 5-HTT mRNA and protein expression significantly across the various alleles. When tested against the S, XS11, LG, LA, XL17, and XL18 alleles, respectively, 100 μM berberine increased 5-HTT promoter activities by 67%, 128.7%, 106.9%, 100.4%, 26.2% and 82%, 2 μM evodiamine increased 5-HTT promoter activities by 216.7%, 81.6%, 305.6%, 181.5%, 175.3% and 102.2%. Berberine and evodiamine increased 5-HTT promoter activity differently depending on the genetic variation of the 5-HTTLPR polymorphism. This study has provided a convincing example of how herbal compounds influence the expression of one of the most intensively studied psychiatric candidate genes, the serotonin transporter."

"Potential influence on sexuality and erectile dysfunction
J Chem Biol. 2012. Effect of novel synthetic evodiamine analogue on sexual behavior in male rats. Department of Pharmacology, AISSMS College of Pharmacy, Kennedy Road, Near R.T.O, Pune, 411 001 India.AbstractCurrently phosphodiestrase5 (PDE5) inhibitors are the first-line treatment for erectile dysfunction. Drugs such as sildenafil and tadalafil are available as PDE5 inhibitors which are potent and reversible but lack selectivity with side effects such as headache, facial flushing, dyspepsia, and visual disturbances. We herein report for the first time novel condensed thienopyrimidines as evodiamine analogue and their effect on sexual behavior in male rats hitherto unreported. Novel synthetic evodiamine significantly showed improvement in male rat copulatory behavior.

Pharmacological profile of evodiamine in isolated rabbit corpus cavernosum.
Eur J Pharmacol. 2002.
This study was designed to examine the pharmacological properties of evodiamine in isolated rabbit corpus cavernosum. In phenylephrine-precontracted cavernosal strips, evodiamine induced a concentration-dependent relaxation. In summary, evodiamine possesses a potent corporal relaxing effect which is attributable to endothelium-independent properties probably linked to charybdotoxin-sensitive K(+) channel activation in the cavernosal vasculature and by nonselective interfering phosphodiesterase to prevent cyclic nucleotide degradation. Furthermore, the physiological effects of evodiamine on the aged animals may implicate a potential for the treatment of erectile dysfunction."

I know that there was tested Berberine, and seems like without success and with lot of side effects. As we can see from this study, evodiamine is more effective for increasing SERT expression. More then this i don't see any side effect of evodiamine other than increased temperature.
Anybody who can buy it , we should test it.

One thing that i can't find is the dosage.

So I think we better try this before trying the drug evodiamine.

Here is a study that talks about this.

There is nothing about SERT.

I've tried it, precisely dried fruits of Evodia Rutaecarpa (Wu Zhu Yu) It cleared my derealization substantially in similar way to Bacopa. Worked for 2-3 days. After month(s) of abstinence from it still I feel nothing beneficial. In my case it was sexually neutral in contrary to Berberine.

I haven't took it for long (days only), my urine smelled like Evodia, so I gave it up.

Research is promising though. PM me if looking for a cheap seller.

How much did you take? For how long? Did you feel the ''serotonin boost''? Do you think it stopped working becouse desensitation? I' ve read evodiamine has low absorbtion in the gut and low plasma bioavailabilty.

Around 6-8 grams for 3-5 days. Just stopped working same as Bacopa after 2-3 days. At that point nothing serotonergic, no desensitization. Why? Dont know. First dosage was a blast, those good little brain zaps, etc. Next one was weaker and so on. I think that bioavailability isn't an issue.

Regulatory effects of Wuzhuyutang (Evodiae prescription) and its consisting herbs on TPH2 promoter
https://www.ncbi.nlm.nih.gov/pubmed/19943499

Seems like Evodia and Bacopa are similar, both increase tryptophan hydroxylase activity. Something is missing though.

According to my experiences Rhodiola Rosea is comparable to Berberine in action although Rhodiola feels numb after overloading and Berberine suicidal, "funny" deep chemical depression for 4 hours, interesting experience btw. Both Berberine and Rhodiola increase erection hardness in my case and improve libido for longer, like 4-5 days. Rhodiola regains its magic after 2-3 months of abstinence. Berberine don't know threw it away, same as Bacopa and Evodiamine. I don't have plans to experiment further with that herbs (except Rhodiola). Short term action imo.

Interesting stuff imo is Ginkgo (safe bet), also Ginkgo in combos I believe some herbs could act differently.

I was pleased also with Shilajit, have some 40% Fulvic Acid pills, good stuff imo, at first it increases serotonin, after five days decreases sero/increases dopamine. Going to experiment with that further. Cheap legit stuff, after 3 pills had dilated "serotonin" pupils so it's not a dust.

Next one is aged Ggarlic extract with vit C for nitric oxide and erection hardness. Have suspicion that garlic could be similar to Shilajit, at first increases serotonin, then decreases, I could be wrong though.

Thank you for the infos. As supposed, SERT upregulation by gene promoter's activity is strong linked with TRH2 expression. This happens with bacopa and with evodiamine too. So, as you have described, you can feel serotonin boost feeling and initially improvements until desentitation occurs again. This is maybe why the effects end in few days. The net effect on seotonin is bad.
